In general, I would assume that they don't - but if a supporting character becomes popular enough with the players that you might consider "promoting" them to main character status, you might consider the extra investment. If your game were a Star Trek series, that character would become like O'Brien on TNG, or Rom, Nog, or several other bit characters on DS9 who became something more enduring than the average supporting character.

Another thought is that a new player, starting the campaign could assume the NPC, promoting it to Main Character status. I can see role playing issues with this, but a good role player, new to an area might do very well taking over Master Chief Petty Officer Gav Pryk, a Tellarite of irascible temperament, but a heart of gold.....
